НЗЧ (известные и приобретенные формы Г.), обусловленное аномальным накоплением железа в некоторых тканях, характеризуется гиперпигментацией кожи, циррозом печени, сахарным диабетом и др. симптомами, которые проявляются после 40 лет; передается по аутосомно-рецессивному типу, локус HFE расположен на участке p21.3 хромосомы 6. Г. чаще всего ассоциирован с гомозиготной миссенс-мутацией в гене HFE, приводящей к замене C282Y в кодируемом геном интегральном мембранном белке HLA-I. Описаны также нонсенс-мутация в гене рецептора трансферрина 2 (TFR2), локализованного на хромосомном плече 7q, и в гене HFE2 (1q) неизвестной функции. Последняя мутация ассоциирована с ювенильным Г.
